Description
In many cases, drug substances and active pharmaceutical ingredients (APIs) are formulated or synthesized as salts, combining an organic compound with inorganic ions like sodium (Na⁺), potassium (K⁺), or zinc (Zn²⁺). Traditional analytical workflows often require separate techniques for analyzing these species—reverse-phase HPLC for the organic part and ion chromatography or titration for the inorganic ions.
The Zodiac HST OG mixed-mode column offers a single-step solution. Its integrated reversed-phase and ion-exchange functionality allows simultaneous analysis of both organic and inorganic components using standard HPLC equipment. When paired with UV and ELSD detection, it provides an efficient and accurate method suitable for pharmaceuticals, research, and QC labs.
Detection was performed using Evaporative Light Scattering Detection (ELSD) for non-chromophoric ions and UV at 200 nm for guanidine. The method allows for fast and effective separation of small polar analytes and inorganic cations within a short runtime, making it ideal for quality control and reaction monitoring involving guanidine salts or buffers.
Quantification of Inorganic Ions
Although ELSD is not inherently linear at low concentrations, the Zodiac HST OG column allows robust quantification through:
Linear calibration at higher concentrations, and
Non-linear fitting for low levels of metal ions.
This method demonstrates the power of Zodiac HST OG for single-run quantification of metal counterions and organic APIs. By eliminating the need for separate techniques, this approach:
Reduces analytical time and costs,
Enhances productivity in R&D and QC environments,
Improves reliability with both ELSD and UV detection
