Dextromethorphan, a common active ingredient in over-the-counter cough suppressants, can be effectively analyzed using HPLC on the Zodiac HST B2 mixed-mode column. This column features reversed-phase retention along with embedded basic ion-pairing functionality, enabling precise control over analyte elution.

In a simple mobile phase composed of water, acetonitrile (MeCN/ACN), and trifluoroacetic acid (TFA) as the buffer, the elution order of dextromethorphan and bromide can be reversed simply by adjusting the buffer concentration. This flexible retention behavior is particularly useful for fine-tuning separations in method development.

Detection is carried out using UV at 210 nm, making the method suitable for routine analysis of dextromethorphan in pharmaceutical formulations.
