Diazacon (20,25-diazacholesterol) is a cholesterol analog that inhibits cholesterol synthesis and blocks steroid hormone production. It is also used as a bird feed additive to control avian populations through contraceptive effects.

An LC/MS-compatible method was developed using the Zodiac HST B2 mixed-mode column. This method enables the quantitative analysis of diazacon in both biofluids and bird feed samples. Diazacon is retained using a reversed-phase cation-exclusion mode, where the retention behavior is governed by the acetonitrile content and buffer concentration in the mobile phase.

Due to the ion-exclusion mechanism, buffer concentration has an inverse effect on retention—higher buffer concentrations lead to longer retention times. To enhance peak shape and reduce analysis time, a reversed buffer gradient is employed.

This method is also suitable for analyzing other hydrophobic basic compounds and is fully compatible with ELSD and LC/MS detection techniques.
