Mixed-mode chromatography on the Zodiac HST C trimodal column enables the separation of hydrophobic and hydrophilic drugs in a single HPLC method. By combining retention mechanisms, selectivity can be adjusted by varying acetonitrile content, buffer concentration, and pH. Seven common drugs are separated using a gradient method, offering alternative selectivity to traditional reversed-phase chromatography. Optimal retention for basic compounds occurs at pH 3–5, with extended retention at higher pH. This method delivers improved retention and peak shapes for both basic and acidic analytes, making it ideal for drug and pharmaceutical analysis.
