The Zodiac HST RP column employs a unique stationary phase comprising dual functional groups:

  • Anion-Exchange Group: Enables separation of negatively charged molecules.
  • Long Hydrophobic Alkyl Chain: Retains hydrophobic molecules through reverse-phase mechanisms.

At pH ~3.0, most proteins in plasma become positively charged and are excluded from interaction with the stationary phase. This ensures clean separations and protects the column from degradation.

Simplified Protein Management:

Proteins and peptides elute as a sharp peak in void or pre-void time, which can be diverted to waste using a bypass valve during the first 40 seconds for a 50 mm column (time may vary for longer columns).
