Mixed-Mode Chromatographic Separation of GDP-Mannose and GDP-Fucose

Two structurally related nucleotides, guanosine diphosphate mannose (GDP-mannose) and guanosine diphosphate fucose (GDP-fucose), were successfully separated using the Zodiac HST SB mixed-mode HPLC column under LC/MS-compatible conditions. The method enables both analysis and isolation of these compounds. Retention is governed by a combination of weak reversed-phase and strong anion-exchange interactions. Fine-tuning of retention and selectivity is achieved by adjusting buffer concentration and pH. Both analytes show strong interaction with the stationary phase, allowing for effective retention and high-resolution separation.
